    为优化花青素纯化工艺、比较不同黑稻品种中黑米和谷壳花青素产量的高低及其抗氧化性强弱,本研究通过单因素实验探究树脂吸附法中各因素对黑米花青素纯化效果的影响,然后对不同品种黑米和谷壳的花青素纯化后比较其产量,最后用1,1-二苯基-2-三硝基苯肼(DPPH)法比较其抗氧化活性。结果表明,静态吸附平衡时间为4 h,解吸时间为1.5 h,吸附液pH为2.5,温度为30℃,70%的乙醇洗脱时纯化效果最佳;其中,黑米中花青素产量最高的是辐黑香糯(21.3 mg/100g),谷壳中花青素产量最高的是固城黑糯(22.6 mg/100g),且固城黑糯黑米和谷壳的总花青素产量最高(43.2 mg/100g)。谷壳花青素抗氧化能力较黑米花青素提高了13.9%。在对黑稻品种的花青素合成途径的关键基因鉴定时,发现矮血糯中无黄酮-3’-氢化酶基因(OsF3’H),而其谷壳中的花青素产量也最少。这些发现在提升黑稻品种的综合利用价值方面具有更多的理论意义。

    Abstract:

    In order to optimize anthocyanin purification process, comparing the yield and antioxidant activity of anthocyanin in rice grain and husk among different black rice varieties, a single factor experiment design was used to investigated the effect of various factors of resin adsorption in the purification of black rice anthocyanin; Then, the yield of purified anthocyanin of black rice and husk in different varieties was compared; Finally, the antioxidant activity of anthocyanin from different varieties was compared by 1, 1-diphenyl -2- trinitrophenylhydrazine (DPPH) method. The results showed that the optimum purification condition for anthocyanin was 4 h of static adsorption equilibrium, 1.5 h of the desorption time, 2.5 of pH, 30℃ of temperature, 70% ethanol of eluent. The highest yield of anthocyanin in grain came from Fuheixiangnuo (21.3 mg/100 g), however, to husk from Guchengheinuo (22.6 mg/100 g). Meanwhile, Guchengheinuo possessed the highest total yield of anthocyanin, 43.2 mg/100 g. The antioxidant capacity of anthocyanin from husk increased by 13.9% compared with that from grain. In the identification of the key genes of anthocyanin synthesis pathway of black rice varieties, it was found that there was no flavonoid -3 "-hydrogenase gene (OsF3 "H) in Aixuenuo and its anthocyanin yield in rice husk was also the lowest. These findings have more theoretical significance in enhancing the comprehensive utilization value of black rice varieties.
